Amaranthus spinosus is a herb which belongs to the Amaranthaceae family and used by ethnic people in treatment of various infections ailments. As result its medicinal properties, it gaining attention many researchers.
 This research work was carried out isolate compounds present leaves spinosus, characterize, use obtained data validate bioactivity.
 The crude extract using methanol Soxhlet extractor. pure extracts were isolated from column chromatography further purification done Sephadex. structures isolates elucidated nuclear magnetic Resonance Spectroscopy.
 In validation bioactivity plant, some pathogens used, including Escherichia coli, Bacillus species Pseudomonas aeruginosa, Candida albicans Aspergillus niger.
 antimicrobial analysis two plant indicated following zones inhibition (mm) for first isolate: 16.00 ± 0.01, ±0.01, 15.00 0.08, 16.00±0.08, 17.00 0.08. For second isolate, diameter zone includes 6.00±0.08, 5.00± 0.10, 26.00± 5.00±0.08, 16.00±0.10. These results compared with standard antibiotic, ciprofloxacin, antifungal, ketoconazole.
 minimum inhibitory concentration (MIC), bactericidal (MBC) fungicidal (MFC) determined those drugs.
